Ready-to-use Research-Grade HLA Class II knockout iPSCs made using REPROCELL’s RNA Reprogramming Technology.
These cells feature all the advantages of our RNA reprogramming technology, including no retention of reprogramming factors and easy culture in NutriStem hPSC XF Medium. They have been engineered using OpenCRISPR-1 to contain a CIITA (HLA Class II)  knockout mutation, which is carefully characterized and offers significant time savings compared to creating your own reprogrammed and edited cell lines.

The Role of B2M and CIITA.

In humans, the B2M protein is required for the presentation of HLA proteins A-G on the cell surface. Disruption of the B2M gene reduces T-cell activation and interferes with the T-cell-mediated immune response through inhibition of Class I HLA signaling. CIITA regulates the expression of the HLA Class II complex. Knocking out CIITA leads to reduced activity of HLA Class II and suppression of the immune response.
