StemEdit Human iPSC non-HLA class 1/2 (B2M/CIITA Homo double KO) cells were cultured in NutriStem hPSC XF Medium(01-0005) on iMatrix-511 (NP892-011) and stained with pluripotency markers. Robust expression of Nanog, Oct3/4, and SSEA4 confirmed pluripotency.

Analysis of HLA Class I expression. HLA Class I expression (Red Curve) is measured by flow cytometry. Both the B2M Mutant (middle panel) and the Double Mutant (right panel) show strong suppression of HLA Class I expression compared to the Wild Type cells (left panel).


The Role of B2M and CIITA.

In humans, the B2M protein is required for the presentation of HLA proteins A-G on the cell surface. Disruption of the B2M gene reduces T-cell activation and interferes with the T-cell-mediated immune response through inhibition of Class I HLA signaling. CIITA regulates the expression of the HLA Class II complex. Knocking out CIITA leads to reduced activity of HLA Class II and suppression of the immune response.
