Definition of CRISPR-SNIPER

Four Examples of CRISPR-SNIPER in Action

1. Knock-out multiple genes in a single experiment

By optimizing multiple guide RNAs, CRISPR-SNIPER can achieve multiplex gene knock-out or knock-in. Up to five distinct genes in a single experiment can be edited, thereby negating the effects of extended passaging and allowing the modification of several pathway components at once. This not only extends the life of your cells, but also increases the reliability of your results.

2. Insert large gene fragments

Where gene editing efficacy normally decreases when your DNA insert exceeds 2 kbp, CRISPR-SNIPER allows the insertion and detection of gene fragments up to 7 kbp in size. Such an advanced gene editing system permits the insertion of reporter genes which can be used to track gene expression.

For example, fluorescence markers can be inserted to monitor the development of your iPSCs or allow the isolation of specific cell types.

3. Generate biallelic modifications

Your disease of interest may display a variable phenotype depending on whether the disease-causing mutation is homozygous or heterozygous. However, it can be challenging to create heterozygous mutants when your gene of interest shares homology with another gene.

The accelerated screening power and high fidelity digital primer/probe component of SNIPER allow the CRISPR-SNIPER system to distinguish between DNA sequences that are similar, enabling the creation of both heterozygous and homozygous mutants.

4. Increase gene editing accuracy

One of the most difficult genome modifications to achieve are single nucleotide polymorphisms (SNPs). Single base-pair changes are responsible for many monogenic diseases but can be tricky to create at a high frequency with conventional gene editing protocols.
