Product Overview

CD437 is a retinoic acid receptor gamma (RARγ)-selective retinoid agonist shown to effectively inhibit growth and induce apoptosis of a variety of cancer cells1. Two forms of dose-dependent action have been reported for this molecule. One mechanism enables induction of apoptosis by a receptor-independent pathway and the other is regulated by a receptor-dependent pathway and is shown to suppress cell differentiation2. More recently, Liu, et al. demonstrated the significance of retinoic acid (RA) signaling in reprogramming somatic cells to induced pluripotent stem (iPS) cells. Treatment of mouse embryonic fibroblasts (MEFs) with CD437 early in the reprogramming process significantly increases reprogramming efficiency up to 20 fold3. This suggests promoting RA signaling with CD437 may have important implications in reprogramming to a ground-state pluripotency (naïve stem cells).

Solubility
For a 10 mM concentrated stock solution of CD437, reconstitute the compound by adding 1.25 ml of DMSO to the entire contents of the vial. If precipitate is observed, warm the solution to 37°C for 2 to 5 minutes. For cell culture, the media should be prewarmed prior to adding the reconstituted compound. Note: for most cells, the maximum tolerance to DMSO is less than 0.5%. This molecule is soluble in DMSO at 100 mM.
Storage and Stability
Store powder at either 4°C or -20°C, protected from light. Following reconstitution, store aliquots at -20°C. Stock solutions are stable for 6 months when stored as directed.
Quality Control
The purity of CD437 was determined by HPLC analysis. The accurate mass was determined by mass spectrometry. Cellular toxicity of CD437 was tested on mouse embryonic stem cells.
