AM580 is a highly specific small molecule agonist for retinoic acid receptor α (RARα). Retinoids are known for their affect on a number of pathways including those related to cell growth, differentiation, and apoptosis1. Specifically, AM580 has been shown to induce differentiation in leukemic cell lines and inhibit angiogenesis in chick embryos2,3. More recently, Liu et al. demonstrated the significance of retinoic acid (RA) signaling in reprogramming somatic cells to induced pluripotent stem (iPS) cells. Treatment of mouse embryonic fibroblasts (MEFs) with AM580 early in the reprogramming process significantly increases reprogramming efficiency up to 100 fold4. This suggests promoting RA signaling with AM580 may have important implications in reprogramming to a ground-state pluripotency (naive stem cells).

For a 10 mM concentrated stock solution of AM580, reconstitute the compound by adding 1.42 mL of DMSO to the entire contents of the vial. If precipitate is observed, warm the solution to 37 °C for 2 to 5 minutes. For cell culture, the media should be prewarmed prior to adding the reconstituted compound. Note: for most cells, the maximum tolerance to DMSO is less than 0.5%. This molecule is soluble in DMSO at 50 mM.
Storage and Stability
Store powder at either 4 °C or −20 °C, protected from light. Information about stability of Stemolecules in solution is not available. As a general rule, we recommend following reconstitution, store aliquots at −20 °C. Stability of stock solutions should be validated in each application.
The purity of AM580 was determined by HPLC analysis. The accurate mass was determined by mass spectrometry. Cellular toxicity of AM580 was tested on mouse embryonic stem cells.
