Overview

ReproCryo DMSO Free™ is an optimized freezing medium for ES/iPS cells without the component of DMSO, which is based on the never- before innovation by Institute for Frontier Medical Sciences at Kyoto University.
Patent has been granted (Japan).

Data

1. Viability

The following are data of cell viability, showing that the product, ReproCryo DMSO Free™, is competitive with other DMSO- containing freezing media.

■Data: average + SD (n=3)
■Cell line: hiPSC 201B7
■Culture condition: ・Media : ReproXF (feeder dependent) ・Coating: Vitronectin
■Freezing method: slow freeze protocol (move into liquid nitrogen after 3 hours at -80℃)
※Please follow the protocol of each product that applied in your freezing- thawing procedure.

 

2. Recovery condition

The growth condition 3 days after thawing showed that recovery of cells in ReproCryo DMSO Free™ is relatively higher compared with other DMSO-containing products.

3. Pluripotency
【 Immunostaining 】

The pluripotency of thawed iPSC was confirmed by immunostaining with pluripotency markers.

・Immunostaining ・ES cell characterization kit (Millipore, Cat SCR001) ・Nanog: Reprocell RCAB0004PF
・Alexa Flour 488 Anti- mouse IgG (Invitrogen A11029) ・Alexa Flour 488 Anti- mouse IgM (Invitrogen A21442)
・Scale bar: 100 μm

■Culture condition: ・Media : ReproXF (feeder dependent) ・Coating: Vitronectin

【 FACS analysis 】

The pluripotency of thawed iPSC was confirmed with FACS (fluorescence activated cell sorting) immunostained by pluripotency markers.
