High-throughput screening (HTS) has become a powerful procedure in the pharmaceutical industry for a variety of applications. Instrumental improvements in recent years have contributed significantly to this development. However, we realize biological improvements still remaining.

To reach beyond HTS, ReproCELL provides several categories of cells.

A lot of hit-compounds including false positive, indicating poor-specificity and faint activity are still in your lab after HTS. Our cells can be applied to HTS, Hit-to-Lead, and Hit-optimization to obtain true hit compounds quickly and more reliable.

ES cells, Functional cells derived from Primate ES cells

Cardiomyocyte

QT prolongation assay using cardiomyocyte derived from primate ES cells : Cell-based assay is used routinely in drug screening. Since 1990' cell-based assay using recombinant (or not) cell lines has been developed for High-throughput screening. However, cells used in drug discovery are abnormal cells which sometimes have huge differences from normal cells in this case. On the other hand, researchers working for drug discovery are frequently using primary cultured cells which are thought relatively native cells. Disadvantages of primary cultured cells are difficulty of large scale experiments and making reproducibility.
If you use ES cells, you have both native cells and pharmaceutical scale cell based assay.
Cardiomyocyte derived from monkey ES cells are on electrodes of MEA 60 or QT96 system (MEA systems, http://www.multichannelsystems.com/) in Fig.1A and 2) . ECG-like signal from cells can be obtained as shown in Fig.1A.
Furthermore ReproCELL's special plate for QT screening using cardiomyocyte is shown Fig.3. ReproCELL is developing this system for high-throughput use in early through late-stage drug discovery. For this purpose only one beating cardiomyocyte-cluster must be precisely and quickly placed on each electrode. If you use our special plate, you can make 96-format cardiomyocyte array for very short time easily. We are planning to operate large scale(=high throughput) screening of QT prolongation. We have already obtained good signal(s) from this plate which cells are just added.
This system can test many compounds easily. For example, treatment with E4031 to cardiomyocetes, QT prolongation can be detected as shown in Fig.4 (a inverted triangle reperesents "T"-like)
