Anti-HLA antibody test
An anti-HLA antibody is known to be produced when heterogenous HLA enters the human body such as in the case of pregnancy, organ transplant and platelet infusion.
If a recipient has an anti-HLA antibody against a donor HLA antigen at the time of blood transfusion, the efficacy of the blood infusion is seriously compromised due to incompatibility for platelet infusion. In the case of kidney transplants, the graft is rejected immediately due to super-acute rejection.
Accordingly, the screening to check for the presence of an anti-HLA antibody in a recipient is an essential part of transplants and blood infusions.
The widely-used anti-HLA antibody tests include the Lymphocyte Cytotoxicity Test (LCT) and Anti-Human Globulin-Lymphocyte Toxicity Test (AHG-LCT).
However, because of the poor sensitivity, these methods have failed to detect a small amount of anti-HLA antibodies in the specimen.
Our anti-HLA antibody test using "FlowPRA Screening" and "FlowPRA Single Antigen" (distributor: VERITAS; manufacturer: ONE LAMBDA) has superb sensitivity, which enables us to detect low levels of anti-HLA antibodies considered impossible with conventional methods.
Measurement principle

- If an anti-HLA antibody against a specific HLA antigen is present, the HLA antigen binds to the anti-HLA antibody (Reaction (1) ).
- After the reaction (1), rinsing is carried out so that only the anti-HLA antibody complex (anti-HLA antibody + HLA antigen + bead) should remain.
- By adding FITC-marked anti-human IgG, the binding of anti-human IgG with the anti-HLA antibody occurs (Reaction (2)).
- After the reaction (2), rinsing is carried out again. An analysis is carried out after removing the FITC-marked anti-human IgG that is not bound to the complex.
