ReproNeuroDA kit

The cells provided are neural progenitors. After two weeks culture using the attached Maturation Medium dopaminergic neurons will be obtained. The dopaminergic neurons can be used effectively for study of the CNS disease such as Parkinson’s disease.

Expression of dopaminergic neuron markers

Dopaminergic neurons expressing both bIII-tubulin (a neuron marker, shown in green) and TH (a dopaminergic neuron marker, shown in red) are obtained after a 14-day culture.

Dopamine release by dopaminergic neurons derived from human iPS cells

The dopamine release from the differentiated neuron derived from human iPS cells was demonstrated, since dopamine concentration in the culture medium was increased by depolarization (elevate the KCl concentration to 56 mM).

Dopamine uptake by dopaminergic neurons derived from human iPS cells
The differentiated neuron derived from human iPS cells were treated with DMSO or DAT (Dopamine Transporter) inhibitor (GBR12909, 20 µM). Dopamine uptake was measured by Neurotransmitter transporter uptake assay kit (Molecular Device). The fluorescence intensity in control was reduce by DAT inhibitor treatment. That results indicate functional expression of the neurotransmitter transporter.
