Constantly available human hepatocytes with robust enzymatic activity

ReproCELL is finalizing the development of ReproHepato – constantly available, high-quality, and affordable human hepatocytes derived from iPS cells.
A series of ReproHepato products optimized for analysis of a wide variety of hepatic functions including CYP isozymes will be launched.

  • CYP3A4 positive human hepatocytes derived from iPS cells
  • Robust CYP3A4 activity comparable to that of primary hepatocytes
  • Convenient cryopreserved form
  • Constant availability with small lot-to-lot variation
  • Products with high inducibility of CYP1A2/2C9/2C19 and high CYP2D6 activity will also be developed soon

CYP3A4 positive cells derived from human iPS cells

ReproHepato cells have typical human hepatocyte-like morphology and characteristics.The expression of key hepatic components such as CYP3A4 and albumin in a high percentage of ReproHepato cells is indicative of the high purity of these cells.

CYP3A4 induction assay

CYP3A4 activity is inhibited by ketoconazole and induced by rifampicin in a dose-dependent manner, as in the case of primary hepatocytes.

CYP3A4 activity of ReproHepato cells is regulated by the known drugs in a dope-dependent manner. Assay can be performed by using commonly-used assay systems. In the example here, assay was performed with P450-GloTM Assays (Promega) and luminescence measurement system FDSS μCELL (Hamamatsu).
Schedule for ReproHepato product launch

ReproHepato is planned to be launched in Q2 2012. Now ReproCELL is inviting limited number of customers for beta-test, where the customer can evaluate ReproHepato before product launch. Please inquire for the details.

2011 Q4 Accepting beta tests for limited number of customers. Beta test is a free, small scale assay service in which the customer send the compounds to ReproCELL, ReproCELL performs CYP3A4 induction assay and return the results to the customer.
